Technical Basis
A review of available sources identified the following: A human oral LDLo, rat oral LD50, and dog oral LD50 and LDLo toxicity data. Data sources were selected according to the hierarchy listed in Table 3.1. Toxicity data were selected according to type of exposure, species, and route according to the hierarchies stated in Sections 3.3.1, 3.3.2, and evaluated using factors in Tables 3.2, 3.3, and 3.4. The dog oral LD50 was selected for the TEEL-3 basis, as the source is more verifiable and more recent (1981) than the human oral LDLo data from 1933. The dose was reported as a range (20-30 mg/kg), therefore the lower value (20 mg/kg) was used. TEEL-1 and TEEL-2 values were calculated according to Table 3.5.
• TEEL-1 = TEEL-2/11
• TEEL-2 = TEEL-3/6
• TEEL-3 = Dog oral LD50 (20-30 mg/kg)
This review generated TEEL values that are more conservative and therefore inconsistent with Rev. 29A. The TEEL-3 in Rev. 29A was based on dog oral LDLo toxicity data, and while the species and route are the same in the current review, a more preferred data source, an LD50, was selected as the basis. Furthermore, in Rev. 29A the TEEL-2 basis was a rat oral TDLo, however that data was not found in the current review and instead the fill gap adjustment factors were used to calculate the TEEL-1 and TEEL-2 values.
