Technical Basis
A review of available sources identified acute toxicity (LD50, LDLo, TDLo) for several species (human, monkey, dog) and routes. Additionally, AEGLs for nitrogen mustard-2 (C5H11Cl2N, CASRN: 51-75-2) were considered to estimate an appropriate magnitude for TEELs. Data sources were selected according to the hierarchy listed in Table 3.1. Toxicity data were selected according to type of exposure, species, and route according to the hierarchies stated in Sections 3.3.1, 3.3.2, and evaluated using factors in Tables 3.2, 3.3, and 3.4. The most preferred species acute toxicity data, a monkey intravenous LDLo, was selected as the TEEL-3 basis. The human (male, assumed) intravenous TDLo was evaluated as a TEEL-2 basis; however, it was not selected because 1) the record was incomplete (no sex reported), and 2) the resultant TEEL-2 was 4x greater than the TEEL-3, and 3) the AEGL-2 for nitrogen mustard-2 is 0.022 mg/m3, indicating this human dose TEEL-2 (0.48 mg/m3) is not sufficiently conservative. Instead, TEEL-2 and TEEL-1 values were derived according to Table 3.5.

This review generated TEEL values inconsistent with Rev. 29A. The Rev. 29A TEEL-3 basis was dog intravenous LD50, while the current basis is a more preferred species monkey intravenous LDLo (Section 3.3.2, monkey > dog). Additionally, the LDLo ¬ source is more current that the LD50 source (Section 3.3.1, 1965 vs. 1947). The magnitude of current TEELs were determined appropriate by comparison to AEGLs for a similar chemical, nitrogen mustard-2.
